What is PT-141?

PT-141 10mg, also called bremelanotide, is a ring of seven amino acids made in the lab, whose trials measured sexual desire. Its record holds both large trials and a sharp critique of them.

United States regulators approved bremelanotide in 2019 as a prescription medicine for premenopausal women with low sexual desire. The research-grade vial sold here is a different product and has no approval for use in people.

Its nearest relative in this catalogue is MT-2. Where some compounds act on blood flow, PT-141 acts through receptors in the brain.

Its human research has travelled further than most in this catalogue, from small early studies all the way to a pair of large phase 3 trials, both in women.

What the 4 PubMed papers cited for PT-141 looked at, 2006 to 2024

In a 2006 crossover study, 18 premenopausal women with low arousal each had bremelanotide on one occasion and placebo on another, and rated their desire.

The two phase 3 trials were later pooled in a 2022 analysis covering 1,202 premenopausal women with low desire, followed for 24 weeks against placebo, with desire scores and distress as the measures. A 2019 drug profile documented the American approval that followed, for the same group of women.

A 2024 review of the phase 3 data pushed back on how those trials were scored, questioning several of the scoring tools.

The trials and the critique belong together; neither tells the whole story on its own.

Reading the PT-141 record

PT-141 offers a clear lesson in reading trials. The phase 3 programme was large, and it measured desire scores and distress. A later review questioned several of the scoring tools used.

Both readings rest on the same data. The difference lies in how a measured change should be judged. This page gives both, because a reader needs both to weigh the record.

The 2006 study had eighteen women before menopause with low arousal. Each had both bremelanotide and a placebo, one after the other, so every woman served as her own comparison. The two large phase 3 trials are discussed in the 2024 review cited here.
